Instrumentation

Equipment Available in the Collaborative Mass Spectrometry Core Facility:

The CMSC facility Utilizes three mass spectrometers which can be configured for traditional or capillary/microflow LC-MS, direct infusion with ESI or nanoESI, or Liquid Extraction Surface Analysis (LESA) experiments:

  • Ultra-high resolution / accurate mass Thermo Scientific model LTQ Orbitrap Velos mass spectrometer equipped with Electron Transfer Dissociation (ETD) and Higher-Energy Collisional Dissociation (HCD) cell,
  • Thermo Scientific model TSQ Vantage triple quadrupole equipped with ESI
  • Thermo Scientific model TSQ Quantum Ultra triple quadrupole equipped with ESI

We have the following Liquid Chromatography systems, which can be configured for capillary/microflow or standard HPLC flow rates:

  • Agilent model 1260 Infinity capillary HPLC system with autosampler and 1260 Infinity Chip Cube MS Interface
  • Michrom Paradigm MS4 MDLC capillary HPLC system equipped with autosampler and bio-compatible quaternary high pressure pumping system
  • Shimadzu Prominence HPLC equipped with two LC-20AD HPLC pumps, autosampler, and vacuum degasser.

These systems are routinely interfaced with any of the ESI enabled mass spectrometers in the laboratory for on-line LC-MS and MS/MS.

Additional Detectors and Equipment:

Additional modules that may be coupled to the Shimadzu Prominence HPLC include a dual wavelength UV-VIS detector, an evaporative light scattering detector, a ?-Ram detector, and a fraction collector.

Nano-Electrospray Ionization Sources:

Two Advion Nanomate Triversa automated nanoESI sources are available for coupling to each of the ESI enabled mass spectrometers, one of which is equipped with Liquid Extraction Surface Analysis (LESA) capability. Additionally, for microflow/capillary LC-MS applications the CMSC utilizes a Michrom Advance nanoESI source and a New Objective PicoChip source equipped with multiple interchangeable PicoChip columns.

Other Equipment:

Other equipment include multiple fume hoods, analytical balances, centrifuge, speedvacs, vortex machines, sonicator, solvent evaporators, vacuum rotary evaporators, water bath, -80°C freezers, -20°C freezers, combo refrigerator/ -4°C freezers, automatic pipettors, stir/hot plates, an extensive assortment of glassware, spare vacuum hardware components, Swagelok fittings, and electronics components, including high voltage power supplies. An extensive catalog of solvents and chemical reagents are available within the organic synthesis and biological sample preparation labs.

OTHER RESOURCES:

Numerous BSL-2 level cell/tissue culture rooms, equipped with laminar flow hoods and incubators, are available to CMSC personnel through member laboratories of the Department of Physiology at Michigan State University. The Chemistry Department, where the CMSC facility is located, has building-wide high-speed internet via wall and wireless connectivity. The CMSC has access to Departmental or University machine shop, glass blowing, and electronics shop services (all of which are fully staffed and have state of the art capabilities). Michigan State University has subscriptions to research databases such as Web of Science and SciFinder Scholar.
